    Study on the bolt-mesh-anchor support technology for mining roadway in complex coal seam

    The stability of mining roadway is affected significantly by the condition of the surrounding rock and stress regime, which is the key factor for determining the roadway support program. Located in syncline axis, the No.15 seam is a steep seam at - 230 m level in Changgouyu coal mine, the original flexible shield cannot meet the safety and production requirement. The No.15 seam is classified as unstable and relatively complex according to the analytic hierarchy process (AHP). The distribution characteristics of stress in the seam around the synclinal axis before and after mining are analysed by use of FLAC2D. According to the complexity of the coal seam and the stress distribution characteristics, the supporting parameters using bolt-mesh-anchor were selected and implemented successfully in an underground roadway, which can be referred to as in the design of roadway support in similar complicated coal seams

    RefSelect: a reference sequence selection algorithm for planted (l, d) motif search

    Background The planted (l, d) motif search (PMS) is an important yet challenging problem in computational biology. Pattern-driven PMS algorithms usually use k out of t input sequences as reference sequences to generate candidate motifs, and they can find all the (l, d) motifs in the input sequences. However, most of them simply take the first k sequences in the input as reference sequences without elaborate selection processes, and thus they may exhibit sharp fluctuations in running time, especially for large alphabets. Results In this paper, we build the reference sequence selection problem and propose a method named RefSelect to quickly solve it by evaluating the number of candidate motifs for the reference sequences. RefSelect can bring a practical time improvement of the state-of-the-art pattern-driven PMS algorithms. Experimental results show that RefSelect (1) makes the tested algorithms solve the PMS problem steadily in an efficient way, (2) particularly, makes them achieve a speedup of up to about 100Ă— on the protein data, and (3) is also suitable for large data sets which contain hundreds or more sequences. Conclusions The proposed algorithm RefSelect can be used to solve the problem that many pattern-driven PMS algorithms present execution time instability. RefSelect requires a small amount of storage space and is capable of selecting reference sequences efficiently and effectively. Also, the parallel version of RefSelect is provided for handling large data sets

    An Affinity Propagation-Based DNA Motif Discovery Algorithm

    The planted ( , ) motif search (PMS) is one of the fundamental problems in bioinformatics, which plays an important role in locating transcription factor binding sites (TFBSs) in DNA sequences. Nowadays, identifying weak motifs and reducing the effect of local optimum are still important but challenging tasks for motif discovery. To solve the tasks, we propose a new algorithm, APMotif, which first applies the Affinity Propagation (AP) clustering in DNA sequences to produce informative and good candidate motifs and then employs Expectation Maximization (EM) refinement to obtain the optimal motifs from the candidate motifs. Experimental results both on simulated data sets and real biological data sets show that APMotif usually outperforms four other widely used algorithms in terms of high prediction accuracy

    Hydrogeochemical characteristics and genesis of Hongshuilantang Hot Spring and its water temperature anomalies during the Rushan earthquake swarm in Eastern China

    Water temperatures of hot springs close to tectonic fault zones often show some variations before earthquakes, and analyses of earthquake precursors in hot springs have significant referential meaning for earthquake monitoring and forecasting. This study measured the concentration of major ions in water from the Hongshuilantang Hot Spring in 2017 and 2020. The ion composition was classified by hydrochemistry into the HCO3·SO4-Na chemical type. The composition of hydrogen and oxygen isotopes in the Hongshuilantang Hot Spring were located near the global meteoric water line (GMWL), indicating that the recharge source of the hot spring was meteoric water. The δD and δ18O values were not plotted on the Glogal Meteroric Water Line (GMWL), and there were some deviations, which suggested that hot spring water underwent water–rock interactions. Deep circulation water played an important role during the evolution process of thermal water. Water temperature showed a decreasing trend from October 2013 to June 2015 during the Rushan earthquake swarm in eastern China. Because of the occurrence of the earthquake swarm, we inferred that regional stress in this area began to be released, allowing continuous rebalancing. Free surface water appeared in some aquifers, and the seepage of low-temperature underground water into the upper aquifer led to a drop in water temperature in the hot spring. The Hongshuilantang Hot Spring and the epicenter of the Rushan earthquake swarm were located on the Muping–Jimo seismological fault zone, with the same seismotectonic system and some genesis relationships

    Tip60 Suppresses Cholangiocarcinoma Proliferation and Metastasis via PI3k-AKT

    Background/Aims: Aberrant expression of Tip60 is associated with progression in many cancers. However, the role of Tip60 in cancer progression remains contradictory. The aim of this study was to investigate the clinical significance, biological functions and underlying mechanisms of Tip60 deregulation in cholangiocarcinoma (CCA) for the first time. Methods: Quantitative real-time PCR (QRT-PCR), western blotting and immunohistochemistry staining (IHC) were carried out to measure Tip60 expression in CCA tissues and cell lines. Kaplan–Meier analysis and the log-rank test were used for survival analysis. In vitro, cell proliferation was evaluated by flow cytometry and CCK-8, colony formation, and EDU assays. Migration/ invasion was evaluated by trans-well assays. Phosphokinase array was used to confirm the dominant signal regulated by Tip60. Tumor growth and metastasis were demonstrated in vivo using a mouse model. Results: Tip60 was notably downregulated in CCA tissues, which was associated with greater tumor size, venous invasion, and TNM stage. Down-regulation of Tip60 was associated with tumor progression and poorer survival in CCA patients. In vitro and in vivo studies demonstrated that Tip60 suppressed growth and metastasis throughout the progression of CCA. We further identified the PI3K/AKT pathway as a dominant signal of Tip60 and suggested that Tip60 regulated CCA cell proliferation and metastasis via PT3K-AKT pathway. Pearson analysis revealed that PTEN was positively correlated with the Tip60 level in CCA tissues. Conclusion: Tip60, as a tumor suppressor in CCA via the PI3K/AKT pathway, might be a promising therapeutic target or prognostic marker for CCA
